Here is a question/comment for Courtney and the rest of the class:
1. I do not like the quantitative results presented in Figure 2 of Courtney's paper. My opinion grows out of taking the 100% from the perfectly matched primer fluorescence intensity. Why is this assumption not perfect?

You know, come to think of it, I agree. You cannot know with 100% certainty that the only thing that is tagged if the one molecule that you are tracking. There is interference in this method in many ways! When I was reading the paper, I noticed that there is no limit of detection noted, no error measurement made, nothing! It doesn't strike me as completely analytical in nature. You cannot just eliminate all other forms of interference just because you believe that it should be negligible.
